Popis Most of toxicity studies with environmental pollutants focus only on single agents whereas organisms are exposed to complex mixtures in the environment. These mixtures often contain compounds able to activate aryl hydrocarbon receptor (AhR) or act as anti/estrogenic or anti/androgenic chemicals. Between these, most frequent are polychlorinated dioxins and furans, polychlorinated biphenyls, pesticides and also polycyclic aromatic hydrocarbons (PAHs). Environmental mixtures can contain many of these components and analyses of the resulting interactions can be exceedingly complex. Thus, defined mixtures of two or more compounds are widely used to study toxic effects of mixtures. In this study we measured potency of selected PAHs and their binary and ternary mixtures to act through some specific modes of action (such as AhR activity, antiandrogenicity etc.) using in vitro bioassays based on mammalian cell lines or yeast-based assays. Studied PAHs, namely benz[a]anthracene (BaA), benzo[a]pyrene (BaP), dibenz[a,h]anthracene (DB[a,h]A] and fluoranthene (Fl) were tested either individually or as binary or ternary mixtures and obtained results of mixtures were compared with concentration addition model and suitability of yeast assays for PAHs mixture toxicity testing was evaluated.
