Popis The fish non-specific immune response includes cellular (phagocytes) and humoral parts (complement system mainly) or the systemic inflammation. In fish, like in mammals, the stimulation of the phagocyte cell membrane, with accompanying activation of the membrane associated NADPH-oxidase, initiates increased oxygen consumption and the production of reactive oxygen metabolites (ROM) with microbicidal activity in a process termed as the respiratory burst (RB). Production of several ROM, such as the superoxide anion radical (O2-), hydrogen peroxide (H2O2), singlet oxygen (1O2) and the hydroxy radical (OH-), has been reported in fish. The aim of this study was to analyze the phagocytic capacity of common carp (Cyprinus carpio) in peripheral blood leukocytes using the luminol-enhanced chemiluminescence (CL) method. Zymozan A was used as an activator of phagocytic reaction in freshly collected whole blood. After one-hour measurement, the kinetics parameters i.e. peak and integral were evaluated. The samples were collected in five periods of one year including different seasons (early summer, late summer, autumn, winter and spring) and a total of 160 individuals (including both males and females) were used. Blood samples were analysed immediately after collecting and also approx. 3 hours after collection. The spontaneous activation of leukocytes during storage period was clearly observed in all five periods. The seasonal changes in RB were detected in raw data as well as after correction for phagocyte number. The highest CL activity counted for 1000 phagocytes was observed in the autumn period, than the activity decreased to approx. one-half during winter and similar values were obtained also in spring. The lowest values of activity of phagocytes (approx. five-times lower than in autumn) were detected in the summer.
