Popis Wedelolactone is an active polyphenolic coumestan isolated from the plant extracts of Wedelia chinensis and Eclipta prostrata. Recently, wedelolactone has been shown to reduce growth cancer cells. These effects were attributed to the inhibition of NFkB and androgen receptors. In this study, we tested the effect of wedelolactone on proliferation, differentiation and apoptosis of breast cancer MDA-MB-231 cells. Wedelolactone in uM concentrations suppresses growth of MDA-MB-231 cells by blocking cell cycle progression in the S- and G2/M phases. The 10 uM wedelolactone induces cell differentiation and the 30 uM wedelolactone reduces viability and induces apoptosis of MDA-MB-231 cells. We found that the growth inhibitory and proapoptotic effects of wedelolactone do not result from deregulation of NFkB activity but they are more likely to be attributed to its ability to bind dsDNA, to inhibit topoisomerase IIa and to induce DNA damage.
