Popis Polycyclic aromatic hydrocarbons (PAHs) are similar to their N-heterocyclic analogs (N-PAHs) important and widespread environmental contaminants generated especially by the incomplete combustion of organic materials. Many of them are known carcinogens, mutagens and teratogens. Teratogenicity of PAHs and N-PAHs could be caused by disruption of retinoid signaling by these compounds, as retinoid levels are crucial for differentiation process and normal embryonic development. In this study the interference of PAHs and N-PAHs with retinoid signaling system was tested using P19/A15 cell line (Approximately one half of tested compounds downregulated RA activity after 6h exposure with lower or no downregulation after 24 h. After 24 h exposure most of three, four and five ring PAHs and N-PAHs increased RA activity. Finally, P19 cells were exposed to selected active compounds either alone or in co-exposure with RA and effects on differentiation process (analysis of Oct-4 protein levels modulation) were assessed.
