Taxonomic characterisation of Aeromonas hydrophila complex

Description The aim of this study was to clarify taxonomic position of A. salmonicida spp. strains deposited at Czech Collection of Microorganism within the “A. hydrophila” complex. In total 11 strains of A. salmonicida and 6 type strains of A. bestiarum, A. hydrophila subsp. hydrophila, A. hydrophila subsp. dhakensis, A. hydrophila subsp. ranae, A. aquariorum and A. popoffii were characterised using biotyping, whole-cell protein profile analysis, ribotyping and gene sequencing (cnp60). Two unknown isolates of Aeromonas sp. from water were included into the analysis. Results of whole-cell protein analysis distinguished different species of “A. hydrophila” complex into independent clusters. A. salmonicida subsp. masoucida and A. bestiarum were clearly separated from the others strains at the similarity level less than 50%. A. hydrophila subsp. dhakensis and A. aquariorum clustered together with the 86% similarity, which support a theory that these species represent one taxon.Genetic analysis based on ribotyping also separated the strains into independent subclusters of A. bestiarum and A. hydrophila subsp. hydrophila (52% similarity), A. salmonicida subsp. masoucida and A. hydrophila subsp. ranae (64% similarity). A. salmonicida subsp. salmonicida were clustered together with A. salmonicida subsp. achromogenes (84% similarity) and one isolate of Aeromonas sp.Results of ribotyping also showed that A. hydrophila subsp. dhakensis and A. aquariorum are closely related, however the similarity was less than 60%.However further genetic analyses (e.g. gyrB and rpoD gene sequencing) or DNA-DNA hybridization would be necessary to reliable identification and verification of taxonomic position of this complex.
