Description Freeze-fracture technique is method for visualisation membrane surfaces of cell organelles. This method used cryo-fixation for stabilization a sample. Sample is frozen in nitrogen and cut in the chamber under a vacuum and low temperature. Glycerol is used as a cryoprotectant for observation fine and smooth cellular structures. Although, cryoprotectants serve for the substitution water and elimination ice crystals production, they could affect a form of fracture through biological membranes and could affect interpretation of morphological structures.
