Description Substrate-assisted laser desorption (SALD) coupled with ICP MS is technique for elemental analysis of samples deposited on suitable surfaces. Indirect immunochemical analysis with labelling based on the principle of antibody-antigen immunocomplex formation is nowadays analytical method routinely used in both research and clinical practice. The signal corresponding to immunocomplex formation is usually amplified by an enzyme. The current trend is for such analyses miniaturize and possibly determine multiple antigens simultaneously in one reaction volume. In this work, the enzyme was replaced by a nanoparticle and the possibility of single label detection by ICP MS was verified, The reaction was further miniaturized to an area of 800×800 µm. Uncorrupted nanoparticles desorbed by laser ablation are introduced into the ICP MS as a sensitive detector capable of detecting individual nanoparticles.
