ISOLATION AND CULTIVATION OF AEROPHYTIC GREEN ALGAE AT LIMITING CONDITIONS OF LIGHT AND TEMPERATURE

Description | Aerophytic green algae represent a specific group of Chlorophytes. They are usually coccal (Trebouxia) or filamentous (Stichococcus or Desmococcus). The aerophytic species are interesting by its ability to survive in the extreme conditions of light, temperature and humidity. They also can take part in the symbioses as a lichen phycobionts. Although it is quite complicated to find out the relationships between algae growth and humidity, the methodology for the temperature and light testing is available. Usage of crossed gradients of the light and temperature has a long tradition in Czech and Slovak phycology (Lukavský, 1982). This study presents the isolation and cultivation in crossed gradient of light and temperature on aerophytic algae and phycobionts. The aim was to find the physical limits of the growth of several aerophytic species of algae. Used algal cultures were sampled at the field, isolated from lichens and taken from collection. Isolation of the phycobiont from lichen was done by the differential centrifugation method (Drew and Smith, 1967) supplemented with micro-filtration (Yamamoto et al., 1993). As a light-temperature treatment were used serological dishes containing all studied strains. As a cultivation medium was used BBM3N. The serological dish had the eight-replicates of the one treatment and one species. The dishes were arranged to the crossed gradients of temperature and light. The biomass was measured each sampling day by photogrammetry. The classification of intensity of green colour and covering area were the actual parameters of the culture under different conditions. The pictures were evaluated by Lucia for classification. Beside this, the chlorophyll a content was measured by multi-scan Bichromatic spectrophotometer at the beginning and the end of the experiment. The results show that the production of individual species are limited by the combination of the studied factors. |
