Description | p53 tumour suppressor is a transcription factor mediating an adequate response of the cell to various kinds of stress by preventing cell division and/or inducing apoptosis. The p53 gene mutations have been detected in almost 50% of human cancers. The most of p53 aberrations are represented by point missense mutations that are predominantly loca-ted in the central DNA-binding domain. Some mutations do not completely abrogate p53 function. In addition to the classic inactivating mutations, there are also dominant negative, gain-of-function, temperature- and cold-sensitive, discriminating and super-active p53 mutations. Several approaches have been developed for detection and analysis of p53 mutations: first, immunochemical methods designed to detect the p53 protein levels; second, molecular analyses targeting changes in DNA structure and, third, functional assays exploring some of the biological properties of the p53 protein. Functional analysis of separated alleles in yeast (FASAY) analyses the transactivation ability of the p53 protein expressed in yeast cells that have been transformed by the RT-PCR product derived from the mRNA isolated from analysed cells/tissues. This method proved to have capacity to be continuously developed and further improved to become an important tool for distin-guishing various functional types of p53 mutations. |
