Description | In this study we performed analysis of effects induced in BM2 monoblasts by ocadaic acid (OA). OA acts as inhibitor of serine/threonine phosphatases PP1 and PP2A. We found that BM2 monoblasts treated with OA in 10nM concentration arrest growth, increase cell size and develop adherency. At the same time, the cells increase intracellular concentration of vimentin, phagocytic activity and capability of producing reactive oxygen species. In contrast, the cells treated with OA in cncentrations of 20 nM and higher undergo apoptosis as documented by cell morphology, DNA fragmentation and phosphatidylserine translocation to external part of the plasmatic membrane. At the same time, the cells treated with 20 nM OA increase production of the c-Fos protein and decrease production and activity of the v-Myb protein. These results document in lower concentrations, the OA induce monocytic/macrophage differentiation, while inhigher concentrations tha OA induces apoptosis of BM2 cells. |
