p53 induces differentiation but not apoptosis of v-myb-transformed monoblasts

Description | p53 is a tumor suppressor protein that protects cells from transformation. In contrast, the v-Myb oncoprotein of avian myeloblastosis virus (AMV) transforms myelomonocytic cells in vitro and induces acute myeloblastic leukemia in vivo. To examine functional interactions of the v-Myb and p53 proteins, we ectopically expressed human p53 in v-Myb-transformed chicken monoblasts BM2. The p53 protein arrested growth in G2 phase of the cell cycle and induced monocytic differentiation, but it did not cause programmed cell death of BM2 cells. Transcriptional activation by v-Myb was up-regulated by the p53 protein, as previously observed during the phorbol ester-induced differentiation of BM2 cells. These results demonstrate that p53 can alter the function of the v-Myb oncoprotein and thus suppress its transforming effect. |
