Description Retinoic acid (RA) and okadaic aicd (OA), the inhibitor of serine/threonine phosphatases of type 1 and 2A can induce differentiation of various leukemic cell lines. v-Myb-transformed chicken monoblasts BM2 express v-myb, the oncogene that blocks the monocyte/macrophage differentiation pathway. In this study, using analyses of cell morphology, cell cycle, phagocytic activity, production of reactive oxygen radicals, expression of differentiation markers Mo-1 and vimentin we determined that OA can induce differentiation of BM2 cells to monocytes and macrophages in concentration that inhibits activity of PP2A but not PP1. In addition we showed that RA enhances the pro-differentiation effect of OA on BM2 cells.
