Molecular characteristics of Bifidobacterium strains collected in the Czech Collection of Microorganisms (CCM) and Laktoflora(CCDM) and reclassification of Bifidobacterium adolescentis CCM 3761 as Bifidobacterium pseudocatenulatum

Description Sixteen Bifidobacterium strains of eleven species were used to evaluate the discrimination power of the tested methods. The strains were obtained from the Czech Collection of Microorganisms (CCM, Brno, Czech Republic) and Laktoflora (CCDM, Culture Collection of Dairy Microorganisms (Tábor, Czech Republic). DNA was amplified using two different genus-specific Bifidobacterium primer sets targeted on the 16S rDNA region. Amplicons 914 bp and 523 bp were used for the restriction analysis using BamHI, Sau3AI, TaqI, Sau96I, AluI, and NciI endonucleases. Seven random primers and BOXA1R and (GTG)5 primers were used for the evaluation of RAPD and rep-PCR, respectively. When characterizing the studied strains it was found that the results obtained for the strain B. adolescentis CCM 3761 did not correspond with its taxonomic position and the strain should be reclassified as B. pseudocatenulatum. The amplicons of the 16S rDNA region of B. adolescentis CCM 3761 and B. catenulatum DSM 16992T, B. pseudocatenulatum CCM 7094T, and B. adolescentis CCM 4987T were used for sequencing and for confirmation of species identification.
