Description The use of organic solvents in electrophoresis medium is well known tool for adjusting selectivity in capillary electrophoresis (CE). The similar effect stands for using virtually non-aqueous media applied in CE. A wide range of organic solvents with different physico-chemical characteristics offers many alternatives for improving selectivity and efficiency. Further, the production of Joule heat in non-aqueous CE (NACE) conditions is low because of low conductivities of organic electrolytes. Thus low generated currents allow the use of high field strengths. In our study we have focused on the influence of background electrolyte (BGE) properties on separation of DXM and its metabolites by NACE. Three organic solvents of different characteristics have been chosen for BGE containing ammonium acetate (10 and 40 mM), namely methanol, acetonitrile and 2-propanol. Separations of the object analytes with the addition of levallorphan (internal standard) were performed in a 75 um I.D. fused-silica capillary, 48.5/40 cm Ltot/Leff, respectively. The evaluation of the effect of two acetate concentrations and change of the apparent pH of BGE in all three solvents was based on the resolution of all analytes, the efficiency of levallorphan separation, electroosmotic mobility and analysis time.
