Description | Non-aqueous capillary electrophoresis (NACE) is based on the use of the electrolyte solutions prepared from pure organic solvents or their mixtures. The use of organic solvents in the background electrolyte enhances hydrophobic analyte solubility and method selectivity. Thanks to high separation selectivity, NACE has gained a renewed interest for separation of drug substances. Dextromethorphan (DEX) is an antagonist at opioid receptors which is commonly used as a treatment against cough and pain. In addition, DEX is well established as a probe drug to determine cytochrome P450 2D6 activity. In our study we have focused on NACE for DEX metabolite determination in presence of parent drug and levallorphan used as internal standard. The effect of ammonium acetate, percentage of acetonitrile and temperature on separation was investigated. The measured responses were migration time of the last compound (IS), satisfactory resolution of a problematic pair (DEX–DTR), peak efficiency of levallorphan and normalized peak area of analytes. |
