To control the quality of RNA biogenesis in the nucleus, cells use sophisticated molecular machines. These machines recognize and degrade not only RNA trimmings-the leftovers of RNA processing-but also improperly processed RNAs that contain defects. Using this mechanism, cells ensure that only high-quality RNAs are engaged in protein synthesis and other cellular processes. One of the RNA quality-control pathways involves the exosome and the Trf4/Air2/Mtr4 polyadenylation (TRAMP) complex. The TRAMP complex stimulates activity of the exosome and recruit its RNA substrates. We aim to study how the TRAMP complex identifies the aberrant RNAs and thus recruits them for the degradation machinery. Here we propose to use multidimensional NMR spectroscopy to determine the three-dimensional structure of several protein-RNA complexes that are involved in RNA quality control apparatus. The project is focused on interactions of the RNA-binding subunits of the TRAMP complex and Trf4 with RNA substrates. The
