Description Alzheimer disease (AD) is the most common type of dementia afflicting especially elderly individuals. One of the candidate genes contributing to its pathology is also angiotensin-converting enzyme (ACE) gene insertion/deletion (I/D) polymorphism. ACE is peptidase that as part of the renin-angiotensin system influenced in regulation of blood pressure. ACE is also involved in Abeta protein degradation. We investigated the relationship between I/D polymorphism of the ACE gene and AD in the czech population. In the case-control study we had 261 AD patients and 223 healthy controls. DNA was isolated from smear of buccal mucosa. PCR products were separated by the electrophoresis on 1,5% agarose gel and genotyped after visualization under UV light. In our sample we found no statistically significant difference in allelic and genotypic frequencies. There is apparently higher frequency of II genotype in AD patients compared to controls (OR=1,37, p=0,18), which is consistent with previously published results from the UK. We presume that by increasing our sample size there should be proved significant association also in the czech population.
