Description Gramnegative bacterium Paracoccus denitrificans is able to grow under broad variety of conditions. Adaptation to altered conditions strongly depends on changes in expression of metabolically active proteins. For study on changes of protein levels in whole-cell extract as well as in bacterial membrane of Paracoccus denitrificans, we have developed methods based on two-dimensional electrophoresis with immobilised pH gradients. Optimization of critical points of both protocols is demonstrated. We have compared effect of anaerobiosis on protein composition of bacterial membrane using optimized method. Several proteins on protein maps were sucessfully identified using MALDI-TOF mass spectrometry.
