Popis Inner surface modification of fused-silica capillaries in capillary zone electrophoresis with an appropriate chemical reagents allow targeted selection of separation system for analytes of interest. Polyethyleneimine (PEI) is polycationic highly branched polymer with positively charged primary, secondary and tertiary amino groups in the structure that can be used as fused-silica capillary modifier. Its irreversible adsorption on the silanol groups of the fused-silica capillary surface eliminates negative charge and excess of charged amino groups cause reversion of electroosmotic flow. In our work, we successfully applied polyethyleneimine coated capillaries in analysis of nucleotides and synthetic pentamers. We studied the influence of particular component of Tris/boric acid/EDTA buffer (TBE) on the PEI coated capillary surface and the effect of hydroxyethylcellulose (HEC) and/or B-cyclodextrin on the nucleotide separation. Finally, we analyzed DNA restricted fragments from Nicotiana tabacum.
