Cytokinins (CKs) are known to act in combination with other plant hormones to regulate diverse developmental processes in plants. The exact molecular mechanisms of CK action in regulation of plant development are the subject of intense current research. While genome expression profiling of CK action has already been initiated by other groups elsewhere, here we propose to analyze proteome dynamics in response to increased CK levels in Arabidopsis under conditions similar to those used in monitoring genome-wide changes in gene expression. This will allow, for the first time, a comparison of the transcriptome and proteome data on highly comparable samples. Increase in CK levels will be achieved by treatment with exogenous CKs and by employing a novel dexamethasone-inducible gene expression system pOp/LhGR. A direct comparison of effects of three distinct CK classes is expected to contribute to elucidation of their individual function(s) in regulation of plant development.
